Zayo-Sigma MacConkey Agar is a selective and differential culture medium used for the isolation and differentiation of Gram-negative enteric bacteria based on lactose fermentation. It is widely utilized in clinical, food, water, and pharmaceutical microbiology laboratories for routine diagnostic and research purposes.
Principle
MacConkey Agar contains bile salts and crystal violet, which inhibit the growth of most Gram-positive organisms, thereby selecting for Gram-negative bacteria. Lactose is included as a fermentable carbohydrate, and neutral red serves as a pH indicator.
Lactose fermenters produce pink to red colonies due to acid production.
Non-lactose fermenters remain colorless or pale.
This differentiation makes it especially useful in the identification of organisms such as Escherichia coli, Klebsiella spp., and Salmonella spp.
